Grapefruit juice

· Severity: Major

· Notes for Consumers: Do not drink grapefruit juice or eat grapefruit while taking this medication. Grapefruit may alter the effects of your medication. Serious side effects may occur. Contact your care team right away if you experience new or worsening side effects.

· Notes for Professionals: Advise patients to avoid grapefruit or grapefruit juice during pexidartinib treatment due to the risk of increased pexidartinib exposure and adverse reactions. Pexidartinib is a CYP3A4 substrate and grapefruit juice is a strong CYP3A4 inhibitor.

High Fat Foods

· Severity: Major

· Notes for Consumers: Do not take this medication with a high-fat meal. Serious side effects, such as serious liver injury, may occur. Contact your care team right away if you experience symptoms of serious liver injury: right upper belly pain, loss of appetite, nausea, light-colored stool, dark yellow or brown urine, yellowing skin or eyes, unusual weakness or fatigue.

· Notes for Professionals: Advise patients to avoid taking pexidartinib with a high-fat meal (55 to 65 g total fat) due to the risk of increased pexidartinib exposure which may increase the risk of adverse effects. Administration of a 400-mg dose with a high-fat meal increased pexidartinib exposure by 100%. Take dosages that include the 125-mg capsule with a low-fat meal (11 to 14 g of total fat).
