• Interaction: Grapefruit juice• Severity: Major• Notes for Consumers: Do not take eat or drink grapefruit/grapefruit juice while taking acalabrutinib without approval from your prescriber. Grapefruit juice may increase side effects from Acalabrutinib including fever or chills, cough, sore throat, pain or trouble passing urine, unusual bleeding or bruising, chest pain, dizziness, fast or irregular heartbeat, palpitations, feeling faint or lightheaded, or breathing problems.• Notes for Professionals: Avoid the concomitant use of acalabrutinib and grapefruit juice; significantly increased acalabrutinib exposure may occur. Acalabrutinib is a CYP3A4 substrate; grapefruit juice is a strong CYP3A4 inhibitor. In healthy subjects, the Cmax and AUC values of acalabrutinib were increased by 3.9-fold and 5.1-fold, respectively, when acalabrutinib was coadministered with another strong inhibitor for 5 days.
